The team that takes possession of the ball (the offense) has four attempts, called downs, to advance the ball 10 yards towards their opponent's (the defense's) end zone. When the offense gains 10 yards, it gets a first down, or another set of four downs to gain 10 yards. If the offense fails to gain a first down (10 yards) after 4 downs, it loses possession of the ball.

It's 1st down and 10. It means that the team on offense has four downs to get ten yards. If they run for 3 yards on 1st down then it would be 2nd and 7.

Its first of 4 downs. If the team does not get 10 yards on 4 downs they have to give up the football to the other team. Most of the time if the team does not get 10 yards on 3 downs they will punt the ball to the other team all depending on how many yards the team needs and what side of the field their on. If there on their side of the field they more than likley will punt the ball to the other team so that team will have further to go to score. I hope this helps.
