Take a bus or fly one of the discount Asian carriers. Rental cars are very expensive and you will probably need to hire a driver from the rental car company due to horrendous traffic.

Really quite simple ... Go via tuas checkpoint ... just follow the road and the signs ... about 20km into malaysia you need to take a left (a big sign there that point to Kuala Lumpur) and then its straight all the way.
Do note - its about 380km to KL or about 4 hours drive so get plenty of rest before the drive and bcos the drive is just a very long and straight road with nothing to look at ... get someone along to talk to or you might fall asleep on the wheels.
If you start to feel tired do take a break at the many rest stop along the way.
Hidden camera by traffic cops can be found along the way esp under the bridges so drive within the speed limit (most parts 110km/hr)

That's pretty simple. Te North -South Highway runs from Johore Bahru to North of Penang. Kuala Lumpur is at the halfway point. Once you get on the highway, there are adequate sign boards to guide you to your destination.
The problem you will face when you enter into the cities - Johore Bahru and Kuala Lumpur.

It's quite easy, especially if you travel via Tuas Checkpoint which will bring you straight on the expressway. On the way, you will be able to see roadsigns like "Kuala Lumpur 50km", and arrows will be shown to guide you the direction.
The most complicating part is when you have reached KL, you will get quite confused, especially if this is your first time driving on the roads of KL. I realised the roadnames all start with "Jalan".. haa..

Do you have a car? If you do, then jump to the next paragraph. If not, you'll have to go rent a car from one of the car rental companies and inform them that you're driving to kuala lumpur. You'll probably have to pay slightly more in terms of insurance premiums.
Now, assume you have a car, there are two ways to get out of Singapore to KL if you drive. First is via the woodlands checkpoint, and second via tuas checkpoint.
To go to KL via the woodlands checkpoint, here's how a simple route... (assuming u're departing from somewhere in town).. try to get into Pan Island Expressway (PIE) Jurong-bound. Once you're on it, look out for Bukit Timah Expressway (BKE) and exit to it.. drive all the way through and you'll reach Woodlands Checkpoint. I assume you'll know the documents that you need to drive through. You'll have to pay a toll fee of S$1.20 at Woodlands checkpoint (using cashcard) and a toll fee of RM2.60 at the Johor Bahru end. After u exit from the immigration lanes, go on to the main road and just drive straight. After about 23-25 km or so, you'll see a sign board that says Kuala Lumpur (North). You'll have to filter left to get into the North South HIghway.
For exiting Singapore via the Tuas Checkpoint, get into the Tuas/Jurong bound lanes of PIE or AYE (Ayer Rajah Expressway). If you are on PIE, look out for the signboards which say Tuas Checkpoint when you're in the Jurong area. If you're on AYE, just go on straight and you'll hit Tuas Checkpoint. Toll fee at Tuas is S$3.70 and at the Malaysian side RM8.40. Once out of the checkpoint, you're already on the 2nd link highway and driving on, you'll hit the north south highway to KL.
Some things to note:
1. There's more traffic over at Woodlands Checkpoint as it is cheaper to get into Malaysia from there. However, during peak periods (5pm-8pm on weekdays, eve of public holidays, public holidays) both exit points experience congestion.
2. At Woodlands checkpoint you only pay toll when leaving Singapore, but at Tuas you pay for leaving and entering Singapore.
3. Total toll charges from Singapore to KL are as follows:
a. Woodlands checkpoint way, enter N-S highway via Skudai: RM43.90
b. Tuas Checkpoint way, enter N-S highway via 2nd link: RM52.40
Both exclude toll charges imposed on the Singapore side.
