You do nothing, it's his problem. He is your best friend but that doesn't mean you own him or his problems.
When I was 10 years old, a family moved in next door to us and the youngest son and I became best friends. He was a very gentle boy and we liked to play all kinds of games from playing house to hockey. I knew him better than anyone.
As we got older, he became interested in me but to me he was my best friend and I was interested in his older brother. We went to the show one time and he held my hand throughout the show but when we went home later and he wanted to kiss me goodnight, I couldn't do it because I liked his brother. After that, we went to different high schools and sort of both went our own ways.
From the time we became interested in dating, everyone would tell me that he was gay and I always stood up for him and said he was not besides my own experience with him, I knew he had some girlfriends. Anyway, to make a long story short, I moved to a different province for 23 years and when I moved back home, I made some inquiries about my friend because I was interested to know how his life had been and guess what? To my surprise, I learned that his gay partner had died of AIDS.
You never know what is going on inside another person's head. Be his friend but let him handle this problem on his own. If people are saying he's not a nice person or anything like that, then stand up for him but leave his sex life out of it you really can not tell for sure.

do you care if a donkey is gay? NO, why? cause you are not sexually attracted to the donkey and could care less.
This other person calling your friend gay might just be attracted to this other guy. Otherwise why would he care so much? Kinda like the marine dad in the movie "American Beauty" so obsessed with "fags", and look at him in the end of the movie.

set up a nice date between your friend and the girl he likes. Maybe you could get them to the same place together and then excuse yourself and well, let nature deal the cards! :) pretty soon, if they really like eachother, the others will notice and they might stop their nonsense.
maybe tell the one who is always saying that this person is gay that it takes one to know one!!

it shouldn't matter whether he is gay or not. the best thing to do is ignore these people. its none of their business ultimately. your friend will probably just get more teasing if he gets defended by a girl, and really, you shouldn't justify what they're saying with any attention whatsoever. live and let live--be a good friend and blow off the ignorant people.

u could stand up 2 him... but that might be not the best thing cause u don't want 2 fight his battles 4 him. u could tell your friend 2 stand up 4 himself! and tell him not 2 let any1 hurt him emotionally or physically!

Tell him to start standing up for himself. People only bully people that let them. Even if he gets beat-up, people will respect him for having self respect. Things will change for him when he decides. You can not do anything it is your friends fight.
