Your eyes and light. Color has wavelength, wavelength determines how far away you can see it. Purple and blue lights during the day would be a disaster! Red is the most visible followed by orange, yellow, and green in that order. So, signs are designed to provide you with the most visible warning possible in ambient light and at the greatest distance. Contrast is also very important. White on red and black on orange or yellow, its eye catching. Shapes are meant to be easily recognizable even when your not paying attention. Squares, triangles, octogons, circles. They all hit you just behind the cerebellum.

Yellow: Warning or caution
White: Regulatory
Green: Directional
Blue: Informational
Brown: Recreational
Red: Stop or Danger

The color scheme is set by the Federal government.

yellow - caution
green - information
red - stop
blue - more information (not as important as green)
white - law (speed limits)

The federal government established a nationwide system so that all states have the same basic signs. There are differences state to state, but none major.
