I can't comment on the Virgin Mary nor would I entertain trying. However, I googled ancient pregnancy tests and found this:

Back then, a woman was assured of a pregnancy when her period didn't show up on time, and she started having "symptoms": ie morning sickness, swelling breasts, increased urination, etc. After about a month's gestation, she could probably feel her uterus (if she knew how to palpatate for it).

Of course, the "heads up" given by the Angel Gabriel was a huuuuuuuge clue, and needed, since Mary had not engaged in sexual activity.

Seems like if a guy had sex with his mother to impregnate her with himself you'd have bigger worries than what type of pregnancy test that sl ut used.
