I have a Pioneer A/V Receiver (VSX-816) and would like to run an Acoustimass 6 setup, 2 outdoor speakers and 2 speakers downstairs. Is this possible?

your receiver has the b side or surround backs witch are amplified it is a 7.1 110 watts per channel if you are using a 5.1 system then you need to get a speaker selector 4 zone or source
out of the b side or surround back side on the back of the receiver
into the speaker selector then the other rooms or outdoor speakers plug into the speaker selector the speaker selector is for impedance on the amount of watts your putting to your speakers so they wont sound bad or blow make sure the speakers can handle the watts. couldn't tell if your receiver has a zone 2 .if it does then make sure you hook up analog audio from all components to here them from zone 2.you might have to set up the other zone in the receives menu.
good luck

Unless your receiver can add for an extra amp, I wouldn't recommend adding the additional four speakers to your system.

You can chain together multiple amps/receivers to power the 2nd and 3rd sets. Use the Line Outs (or Tape Outs) of the master unit, and feed it into the inputs of the 2nd unit, and so on, or feed it to a Line-Level splitter to feed 2 or more units directly.

a) Not all at the same time, no.
b) You would fry your amp if you did.

Bose is not very compatible with other brands (and they like it that way I hate to say), but if it is compatible with your system, add amplifiers to increase speaker count.
