It means that X number of calories out of the total number of calories in one serving come from fat. For example, if there are 240 total calories per serving and 120 are from fat, then 50% of that food's calorie count comes from its fat content.

you get calories from everything food (carbs, sugar, protein, fat, etc..) 1 gram of fat contains 9 calories. If it says calories from fat is 5 then there is less than one gram of fat. Not all fat is bad though. Stay away from saturated fats found in red meats and fried food.
