I'd start looking for a new friend. It sounds as though this one is not really your friend, and in any case has some serious problems if she's off and on all the time. But they are HER problems, not yours, so don't feel you have to hang around and help her sort herself out.
Look for another friend, and I hope you find a good one.

I think I'd try a direct approach. Ask why this person doesn't want to be your best friend anymore. You may be able to clear up a misunderstanding. But if you don't buy their answer or they just won't speak to you, it may be time to move on. You'll also have to decide if you're willing to put up with "on again/off again".
