I would ask them if they would like to leave a message. If you ask, "Can I take a message?" then, sure enough, some dumb@ss is going to say, "I don't know, can you?"

If I do not recognize the voice I will ask "who is calling" if the person they are trying to contact is not around I will ask "Would you like me to take a message and pass it off as soon as I see her/him?" I will usually keep a little log of who called what time and why then hand it off.
