Not sure of an exact number. No doubt that there was more than one. There had to be at least two to form a "watch". Around Matthew 27 mentions a "watch" being set by Pilate.

The same one that says how many magi came to Jesus at his house.

Matthew 27:66 says they posted the guard.
How many people do you need to guard a grave against grave robbers?
The only thing that's clear is that chapter 28 indicates multiple guards. "...some of the guards went into the city...". In normal usage, this would indicate at least three guards: more than one, but not all of them. Maybe there was a normal number of guards to set so Matthew didn't feel a need to specify. Or maybe it just didn't matter.
