Well....really only Florence. Once there, I would go to;
"Giardini Bobboli"
"Galleria Uffizi"
"Duomo/Battistero"
and last but not least eat a nice gelato on the "Ponte Vecchio" and sing along to your favourite classics until 2 AM.
Pity that you would only spend a day in Toscana. Really you need at least a couple of weeks....

I have spent much time in Tuscany, and the top three cities are Florence, Sienna and Pisa. Pisa can be done in a day, Sienna in perhaps two days, and reserve the rest of the trip for Florence. Few people go to Tuscany for the day, but if you truly only have one day, then Florence is the tops.

Florence, Pisa (to say you've seen the Leaning Tower, but the cathedral is more interesting) and almost any vineyard (they're nearly all good).
If time is short, visit Cantina Antinori in Florence to sample the best Tuscan wines, David's statue (the real one) in the Accademia Gallery and then the wonderful Uffizi Gallery. No need to leave Florence.

Florence - it is an amazingly beautiful city. I stayed in a place called Pratalino in the mountains around florence and the locals were very friendly. There were also some good thunderstorms.
Pisa is good for a day trip. There is not much there other than the tower but it is worth going to watch all the japonese posing for photographs.
