Or is it ok to just remove one sparkplug at a time? Or does it not matter. (Of course, I will ground the distributor to coil wire.)

one at a time is fine just crank it 3 or 4 rounds before checking it -they should be within 10percent of each other --some same more but i like em close to get em to have power and fuel economy

A compression test is where you are measuring the compression in a cylinder. Essentially, the build up of atmospheric pressure. Really, it won't matter (or shouldn't) how many plugs you remove. Forget the distributor wire. The test doesn't concern any electrical stuff. You will install the meter into the plug port and turn the engine over to test the compression. If you are not in the tolerance specified you have problems such as rings. Anyway, the engine should not be running during the test.

The correct way to do is,remove all plugs,remove coil lead,hold throttle wide open,let each cylinder compress 3 times for each reading.This is the way to get accurate readings,good luck.

It doesn't matter unless you DO have a cracked head or blown head gasket. In that case, you might want to try them one at a time to see which cylinder has the problem and how bad that problem actually it. Otherwise you can test them and make sure they all have compression by removing all the plugs at once.

just remove sparkplugs as u go, you shouldn't have a problem. not to be rude but if u don't know the answer to that question why are u doing it? yes one at a time. anus is wrong, you can't run the engine while you do compression test.

Frank M is the right way,and ground the coil wire,saves the coil.

remove all the spark plugs, I guess that the engine is 2.4, if I'm not wrong, the compression is about 225#, a little high for a regular engine

No! just one plug... run engine do your test..
from this you should be able to get a good read.
