Say a girl or guy that you've been friends with says they want more than a friendship. Does waiting or making someone wait longer for a reply equate to a negative response most times for you?

Not necessarily because with waiting they are still hopeful of a positive reply. But you have to be delicate about this matter if you are saying no because it may change your friendship. In time if there is still no response the other person will get the picture.
But the disadvantage of saying no immediately is that later if you change your mind, then there might not be a chance.

Some of the answer are really hard for me to choose a best answer, when people go to so much effort to give you a answer. The tougher question if they are to hard for me, I go to another question. I don't want to say thank for the 2 points and leave it at that.

Sure...you feel like they really have to think about it...so maybe that is a good thing..but a quick answer is much more reassuring...no one likes to be led on..
