There is no simple answer, but basically it's like lying. Your lie has to have a ring of truth to it. Suddenly making an unusual bet at the end of a hand is like calling your boss and telling him you are sick when in the background he can hear casino noises. To truly sell a bluff it needs to be believable just like a lie. What most amateur players don't realize when they see a great bluff on TV is that the bluff they saw may have been set up dozens of hands ago with a certain pattern of betting or some table comments.

The only best way is to make it believable and not to have tells that betray what you are trying to do. Also the more people you try to bluff, the less likely you are to succeed. Bluff one or two players at the most, make it believable, be sure that they are weak and are likely to fold. In no limit, don't give them the right price to call you. Practice a lot and study the game.

It all depends on the player you are trying to bluff out of the pot. I like to bluff players who are likely to fold to my bet. If you are playing a maniac who has called down every hand he has had to the river, then i would not try and make a move on him, but if he was someone i thought might lay down a marginal hand to my bet then i would fire out at the pot. Just be careful in how you bluff because sometimes you end up trying to bluff someone with a big hand, and they have no intention to lay it down. I don't bluff into coordinated flops, unless I'm semi-bluffing, it is to likely you will get called down by some one with a pair, it also matters on how many players are in the pot as well, if we are in a four way pot then i would not bluff at all. Your bluff also has to be believable, i know if i raised before the flop i will almost always put out a continuation bet if i miss the flop because it gives me fold equity and it makes it look like i have a strong hand, because i have taken the lead in the betting. So when you bluff it has to be the right time and the right player in order to do it right.

Variance is the key to bluffing. What might be the perfect bet for a bluff one hand will not work if you keep doing it. Mix it up. Bear in mind the quality of the opposition. As other responses have said get the image that you only bet with a good hand, look like your a tight player.
The all in move is invaluable as even if he knows your bluffing hes got no room to rebluff so he can only call with a legitimate hand. But if u keep doing it theyll sit there and check to u with a monster, so key word variance!

well, you could try buying the pot (overbetting), but if they call you on that, you're screwed.

Best way, I've found, is to get a reputation for not bluffing - or for betting a certain way when you have a good hand. Then, when you don't have a good hand, you bet that way still - and they'll assume you're holding pocket aces, or something.
