I believe he first case of incest in the Bible would be found in Genesis. With Cain and Abel as the two brothers - and there being no other peoples outside of Adam's family.
But this question has been an old favorite of hecklers, wanting to demoralize a Christian's perspective on his faith.
But truth be known, the answer is twofold. First, we do not read that Cain found himself a wife in the land of Nod. Rather, it was there that Cain " lay " with his wife, who then gave birth to Enoch ( Genesis 4:17). Secondly, although Cain and Abel were but two brothers initially, there were also to be " other sons and daughters " fathered by Adam in his long life ( Genesis 5:4). Cain would have married one of these daughters, when the necessity demanded some intermarrying at a time when the race had yet to increase.
The Bible does not tell us how many children Adam and Eve had in their lives, but they probably had a great many, for they had a great many grandchildren. Since people lived for more than a hundred years in those times, a single person could have as many as a hundred children. So taking that into account, it is not hard to understand how the population of the world increased so very rapidly.
So Cain was the first incestuous relationship acknowledged.

I am going out on a limb here and assume you mean, who committed incest after it was forbidden in Leviticus.

That would be King Davids son, who raped his half sister, who was exceedingly beautiful by biblical description.
