Jesus is dividing those who truly seek after God from those who are just playing "religion".
He is further clarifying that if the religious folks truly understood the scriptures they had, they would know clearly who He is, the Messiah.
Jesus is letting us know as well, that there are two groups in this world, those who truly follow Him and those who do not.
Pick a side.

when He says you are children of the devil he is speaking to the Cainanites - who were the children of Cain, the first murderer.
Also, there were two types of priests in the Temple at Jesus' time - pharisees and saduccees. The pharisees believed in the afterlife - the sadducces did not and taught there was no afterlife. The sadducces were not the original priests - but were Babylonian priests who joined the the tribes of Judah, Benjamin and Levites when they were freed from captivity in Babylon and allowed to return to Jerusalem. The sadducces came to Jerusalem with them and infected YHVH's Temple - dividing it.

Jesus is talking to the Jews, the people that knew the old testament. They worshiped God with their lips, but their heart was far from Him. Jesus was saying if they truly loved and worshiped God they would know He is the Son of God.
