The causative viral agent for mononucleosis is the Epstein-Barr Virus. Many people have carried it and never get sick.
The virus has been isolated in dogs (2005, Journal of General Virology) however, to date, there is no known transmission (of disease) that occurs between human and dogs. The dogs may have picked it up from human carriers and other vectors.
Wash and general hygiene is a good thing. If bro blows his nose or coughs in a tissue (moisture) then touches a doorknob, flushes the toilet (touches the flush handle), -- stuff that is touched and still moist can transmit the virus. Of course, don't share food and utensils with your brother. If you touch the dog, wash your hands, just to be safe. :)
I hope he heals up fast! Once he's over it, then the virus dries up and you don't have to worry about it. It is not as contagious as a flu or cold, but standard hygiene should help keep you safe.

The owner of the dog needs to know how to "operate" the dog, same way you can get into a car that's in perfect running order but if you don't know how to drive you won't have much luck making the car go anywhere.
