There are at least 1500 different varieties being used for commercial wine making, and at least 2000 known varieties worldwide. New varieties are being created all the time by crossings. Three recent new varieties I have tasted in the past couple of years include include Roobernet (Cabernet Sauvignon/Pinotage), Marselan (Cabernet Sauvignon/Grenache) and Nouvelle (Semillon/Crouchen Blanc).
Anthony Hawkins has a useful glossary of wine varieties here http://www.stratsplace.com/hawkins/wgg.html and I thoroughly recommend Oz Clarke/Margaret Rands beautiful book Encyclopedia of Grapes
