Not neccessarily. That has happened to me on the occasional hot day, if they were feeling too tight or if I stained them and did not have a spare pair in my purse. And there are many other reasons. Relax, do not jump too conclusions and when the time seems right ask her about it.

Confront her on what she is doing with them. Maybe your wife could be taking them off for you. Have you guys been married a long time? She could just be trying to "spark" the relationship again.
If she does give you a lame excuse about why her underwear has gone M.I.A., I would re-evaluate the relationship that you have with your wife and possibly seek a marriage counselor or divorce attorney.

You should be asking her some questions, and unless she had an accident and didn't have a bag handy to bring her panties home in, you should be worried.
