Just put 'of as' between think and tired.
Tired is a state and the opposite is usually 'not tired.'

Well, I don't think the sentence makes sense. But you could always just put it like this - "People tend to think I'm always awake and never tired."
You could also say - "People around me think that I never get tired"
The opposite of tired is alert / awake... i'm sure there's more but i can't think of any others at the moment...

The opposite of tired depends on context in this case it would be energetic but elsewhere it could be refreshed.

The opposite of tired is energized. As far as your sentence is concerned, I'm really not sure what it is you're trying to say. Perhaps "I am full of energy" or "I'm not at all tired", will suffice. If you want something with a little more slang, try "Good to go, Bubba. Let's roll!"
