He's most likely interested in you. Winking is a kind of flirtation gesture, and smiling is a positive and welcoming gesture.
So flirt + welcoming = interested, or in my mind it does.
Also, especially since he doesn't do this towards other women, so you're probably special, and once again... interested in.

Being a male, I think this can mean that he is sexually attracted to you. Heck, lots of males in TV shows wink at girls, and it generally means a feeling of love - or something of the like.
