No, a boy does not lose his virginity after masturbation. A person only loses their virginity after engaging in sexual intercourse with another person.
A girl will know whether a boy has lost his virginity only if you tell her.

Nope, losing your virginity involves penetration in most people's opinion. Masturbation definitely does not affect your virginity a bit! B--- jobs may be a little more grey area but I'd say you are still a virgin if you've given or received a BJ. Same way boys know if I girl has lost her virginity... you don't know unless you ask. For girls, a broken hymen doesn't mean not a virgin. It can be broken by riding a bike or a horse. I don't think of Virginity as a physical attribute. Either you've done it or you haven't and there is no physical characteristic that can prove you are a virgin.
