"whats up" is like saying whats going on with you today. a respone would be. "ah, nothing much" "just working".

I am not a "fella" or a "guy" but I have your answer.
I always respond with ME! Then I give a big smile!
I also work in a big office and everybody is friendly enough to ask "What's Up" Most people give the same answer (like you said). Change your answer, be different. Say ME! You will get a nod, a smile and you will be noticed by the people that are tired of hearing "Uh, OK"
The secret is, you have to mean it. A fake response and a fake smile is noticed immediately!

You can say "Besides gas prices? Not a thing...what's up with you?"
"Nothing at all...what about you?"
"Nothing, can't complain, it won't do any good now will it?" (that's my favorite because no matter what situation you're in, you always get a chuckle or a comment for a possible conversation or just a pat for the day.)
"Can't call it...how's everything on your end?"
I hoped these helped. I work in an informal environment like that so I know what you mean. These are just a few phrases that I say without being katty or getting smart.

You might need to know what "Whats up" means before answering. "Hey man, whats up" is just like saying "Hi, what are you up to". Knowing this much might help you to give a truthful response instead of a rehearsed response.
Be loose, shake the nerves, look the person in the eyes, bring about a slight smile, and answer with your own words. --Good Luck.

You say..." a two letter word describing a direction". They asked what "up" is and you're telling them the definition of "up", if they act confused by the response. (I say this a lot and no one gets it at first , but then a few (sometimes up to a day later) they finally get it, or ask what I mean.

"I am good and you" is just fine.
also: I'm cool...how are you?...Just chillin' and you?
or: make sure you ask it first...or at least almost at the same time....lol.
To your last question....what do I do when someone asks me that? Very original: I just smile- as if I didn't hear the question...and then I ask how's it goin'?
