There are regular bus shuttles from JFK to Port Authority and Penn Station. You need to ask the Information desk at JFK

Well, quickest would be a bus or taxi. Not cheap. Cheapest would be train. Not fastest. I would train from JFK to Penn and save 40 bucks.

The Airtrain will be the fastest just becareful when you change at Jamaica - it's not the nicest station.
You can also take the bus however you never know how long because of traffic.

There should be shuttles or flat rate cabs available. At that time of day, it should be plenty of time.
