Do you mean for a personal computer? If so the answer is AT&T Unix System 5.3. This was a project funded by AT&T and Intel and created the first 32 bit OS. it was available at about the time the 386 shipped. SCO Xenix was derived from this OS.

If you are talking about computers in general, than there were many different computers in the 60's and 70's that were 32 bit and a variety of OS'es and I am not sure which could claim to be "the first".
