Your butt has the largest and strongest muscles in the body, so exercising them by lifting weights will make them grow, just like any other muscle, giving you a bigger butt. The exercises you'd do depends a lot on what you want to achieve (the shape of it) and the type of butt you now have.
For example, squatting makes your butt bigger over all. In other words, it makes your whole butt grow. If you have shape (roundness) but want your butt to grow some, them squads should do the trick. If you need size and shape squads combined with other shaping exercises should be the way to go. Squads for size and the other exercises to give it shape or lift.
The girl that posted before me has it right. Muscle weights more than fat, therefore, if you start lifting weight, throw away the scale. Use a tape measurer or a Body Composition Analyzer. A BCA is a device that sends small electrical currents through your body to determine you body composition, therefore giving you a very accurate idea of how much fat your body is composed of in percentages. I'm sure your have seen them. If you have a heart pacer, do not use one and it's important follow the instructions. If you don't have a BCA, you can buy them cheaply at most department stores. Some start at as low as 25 dollars. I would suggest spending a bit more money and buy a more expensive one. Even though most, including the cheap ones, are pretty accurate, it's just that they'll last longer. Your doctor should have one of professional grade.
If your are not on a weight training program, I suggest that you get on one. Weight lifting is one of the most beneficial forms of exercising for women, even more than for men. That is because women are more likely than men to suffer from osteoporosis as they get older. Back in the late 80's when I was training, women were afraid of lifting weights because they were afraid that they would start looking like men. The fact is that it's impossible for that to happen, unless you take steroids. First, women don't have as much testosterone hormone as men, and second, women have much less muscle mass, specially in the upper body, than men to start with.
I worked as a trainer long time ago when Bally Total Fitness was called Holiday Health Spa. I've always had a butt fetish, so since I've always liked butts, I learned has much as I could about them (exercise wise) and applied my knowledge to my female clients. I’ve seen first hand what weightlifting, combined with other exercises, can do to a women’s or even a man’s physique.
Of course there are ways to shape your butt with out weights, but weights, especially if you need mass, it’s the fastest way to achieve your goals. And it’s not like you have to lift weights all they long either, not unless you want become a professional bodybuilder, but just a bit combined with other activities like cycling or rollerblading, can do wonders for you. Get yourself some padded panties, or just stuff some paper towels down there. Ha ha. Okay seriously, the butt is the biggest muscle in the body, so yes you might be able to make it bigger through exercise depending on wether or not your body type gains muscle mass easily (generally women don't as much as men). Even if it dosen't get bigger, excercise can improve the shape. The best butt excercises include squat thrusts and lunges.

Squats, gradualy adding arm weights, leg curls on a bench, using an excersise ball doing pelvic thrusts while squeezing your glutes all work well. Also , lunges while holding arm weights. Laying on your side and doing leg raises with ankle weights for the outer muscles. Remember, muscle weighs more than fat so if your getting the look you want, dont worry about your weight!

Consult a gymnast, he may tell you how to do excercises that tone the butt.
