Scientists have juggled this question throughout the years and the list of answers is ever-changing. In some circles of science, the ability to make and use tools is a sign of our humanity and, thus, our dominance over the other species on the planet. Recent research has found, however, that there are actually quite a few animals with this ability. Some members of the ape family are actually able to crudely whittle sticks of wood to use to find food (usually termites, ants or grubs).
Once our ability to walk upright was considered a sign of our humanity, but we now know that there are members of the ape family that walk upright (on two legs, or bipedal) 90% of their lives.
It has been said that our ability to create and understand complex languages is a signal to our humanity, although science now realizes that dolphins and some whales have languages as complex (if not more) than our own.
So how do you explain that "spark" of difference/intelligence/soul/etc that makes us unique in the animal world?
Here are a few of my observations on the subject. I believe what makes us "human" is our ability to love, to hate, to feel compassion, to seek to attain goals beyond the "primary directives" of food, shelter and procreation and all the extremes that ability may bring us. It has been researched and noted that, even among "humans", many of the separate races tend to act more "animalistic" than others. Science has found that some races tend to be much more "primal" in their lifestyles than others. Even beyond their separate races, humans can be individually ranked as to their likenesses to the animal kingdom. One example of this is the "territory" issue. We put up fences and walls and borders to "mark" our territory and to keep rivals at a distance. This has never been more obvious than the battles going on in the Middle East to this day for a piece of land that has been fought over for over two thousand years. Think, too, of the amount of time we, as humans, spend thinking of food, looking for food and eating food. We, as a society are so morbidly obese due to the fact that an animalistice desire in us (some more than others) tells us to seek food at all times, less it become scarce. We, whether by God, or by some fluke of evolution, were given dominion over the earth and the animals that share it's habitation with us. Through that inheritance and ownership, we are also entrusted with the care of this planet that supports and feeds us. I believe the knowledge of that responsibility, just being able to realize our power and, subsequently, our weakness in the grand scheme of the universe, is probably the "spark" that separates us, elevates us above the "lesser" animals on the planet, and makes us human.

Having the physical attributes that we have, anthropologists call our kind homosapiens or plain human beings. But if I understand what your really mean, humans have the ability to grow spiritually. I'm not referring to getting more intelligent, apes can, to some degree, get smarter. I'm referring to something internal beyond mere intelligence and conscience.*
*Had to stop for a bit to try to define what I really mean but found I cannot. Sorry. Good simple question, though. You stumped my smart-a.ss mouth.
