I have a 7:00 am flight and want to know what time to leave Manhattan.

Early morning... not too long, probably only 20 minutes. But... with all of the new security measures you should probably get there at least an hour early. I'd leave Times Square at 5:00am to be safe.

Given the heightened state of airport security between 5 and 5:30 AM
20 minute taxi ride to the airport.
5 minutes to collect your luggage.
30 minutes Standing on to have your carefully packed clothes tore out of the suitcase by some TSA employee who could unpack them neatly but is in a hurry.
10 minutes to repack your clothes so the suitcase will close properly.
10 minutes to check your luggage.
5 minutes for the restroom break.
30-60 minutes standing in line to be checked for bomb, drugs, and weapons.
If the weather is bad, call your carrier first.

Probably no more than 45 minutes in the morning. Heavy traffic is going the other way.

If I were you I'd take the D train to 145th. Get off there and catch the M60 bus...it goes straight (well not straight) but it does go to LGA.
Unless you wanna take a cab which is $$$$ wasted while you'll be sitting in traffic.
And please allow for check in time...
