Some guys can be very feminine, and still be with girls because their behavior does not mean they are gay. He might well be if though. If he's your best mate, could you not tell from your friendship if he's leaning towards men?
What you can do is ask him. Or let him be. It might be that he's a bit more feminine in his feminine genes than most men. I know of a guy who has family and is in total love with his wife but he's got a golden touch when it comes to makeup and fashion. He is kinda loud as well. Very over dramatic about things. But he's all man because he had been a player when we were all in highschool and girls talk!
So you never know...some guys know ALL there is to pleasure a woman and still be ultra sensitive themselves.
Try asking him...a question: Does this bother you from your best mate? If it does, you should deffinitely let him know. If it doesn't, accept him as he is.

If he is your best friend than except him as he is. What really matters is how he treats you as a friend. As long as he doesn't disrespect you in no form or fashion or come on to you in any way , i would still remain his friend, obviously you have been friends for some time. True friends are hard to come by in this day and age.

My best male friend often acts like a girl, but he's very gay. Or perhaps transgendered, I wouldn't be surprised. He's still my best friend, since it's his personality (whatever it may be) that makes him my friend.

first off, age matters here.
second, just be his friend if you're cool with that. talk to him, maybe he dosen't realize how he is portraying himself or maybe he does. either way, if he is cool and you are cool, then let it be.
