Well, a vestal refers to a priestess who was chaste and a virgin. I think it's kind of referring to the peace of being almost like a hermit, disciplined in desire - without your mind clogged with wishes. The mind is 'spotless' because this person's kind of separated from the world. Each prayer accepted, each wish resigned - almost like Pope imagines they can talk directly to God without their own baggage getting in the way...
Hope that helps.
(I love that film, too.)

Pope is envying nuns, monks, hermits, ascetics--people who have been able to let go of earthly desires and ambitions and resign themselves to a life of prayer and service. The Vestals served in the temple of the goddess of the hearth, Vesta. Pope seems to imagine a life without mental or spiritual conflict--more of an Eastern idea of spirituality, really--hence the choice of this passage for the title of the film.

It's meaningless. That's what poets do. They're failed writers that jumble together random nonsensical words that they think might be deep and though provoking. If they are so great, then why can't they write a 300 page novel like that? They can't, because it would be nonsensical garbage, that's why they only do a couple of verses.
