You feel a definite push on your back and butt, maybe 40 pounds or so, due to the acceleration down the runway. There can be some thumps and bumps if the runway pavement has any dips in it. When they get to flying speed, they "rotate" and the plane pitches up. Not abruptly, but pretty quickly. You are now pointed about 30 degrees into the air and continuing to accelerate.
They will climb at that steep angle for 4 to 5 minutes until they get to 10,000 feet. At 10,000 feet a little "bing" and the flight attendent announcement will tell you that you can use music and video and computers that you brought with you (if you don't need to unbuckle to get to them). The plane flattens its angle a bit (because they can exceed 250 mph once above 10,000 feet). They will keep climbing for another 10-15 minutes until 31,000 to 35,000 feet. Then, weather permitting, you can move about the cabin and they'll start giving you peanuts and soda.
Nothing about it is uncomfortable (but it is exciting the first few times), it is less jarring than a fast turn in a car because all forces are straight down or backwards. No side-to-side forces, even when the plane is turning. I've been in 40-degree banking turns (to avoid thunderheads in the Midwest or mountains in Alaska) and unless you look out the window, you'd swear you were perfectly vertical.

Taking off is the best bit. It's a bit like sex, lots of anticipation, then action, ending with a burst of excitement as you accelerate down the runway and take off!
Basically, the aeroplane will taxi to the runway. It'll feel weird cos you're in this huge thing which feels like it's going quite fast. Finally it will get to the runway, and if it's not busy will taxi onto it. If you're on a small plane like a 737 or 757, as soon as the plane hit's the runway the captain will put the engines too full throtle. If you're in a 777 or 747 then the captain will line up, put the brakes on and put full throtle on, then release the brakes. Either way, the engines will get proper loud and you will feel this sudden acceleration, The acceleration is quite fast, faster than a normal turbo car but not as fast as a ferarri (not that i'd know).
Anyway, as the plane hurtles down the runway, engines roaring away, the captain will tip the nose and the plane will takeoff. Suddenly you don't hear the ground anymore, and you feel a little quesy. Depending on the Standard Instrument Departure the Captain has to follow, the plane will usually climb quite steeply and you'll be passing clouds in no time. It feels a little strange because it's usually quite noisy and it feels a little unstable.
One last thing, take boiled sweets. As you climb, they depressurise the cabin which means you're hearing will start to become a little strange and every-so-often your ears will pop. This is normal since your at the equivalent alt of 6000 feet (to stop to fuselage exploding out). Boiled sweets or milk tend to help your ears adjust.

Well, the plane accelerates down the runway, so that part feels like a car accelerating really hard. And then when it goes fast enough, the nose points upward and you lift off of the ground. It's quite exhilerating!

Mostly, it's like driving down the street...on a sort of bumpy road.
Then, the plane gets off the ground, and it smooths out a bit.
There's a bit of g-force applied during the lift-off, kinda like if you were on a roller coaster, and a bit of pressure that you can feel in your ears.

It feels great!!!! It's kind of like a car accelerating really fast down the road and you feel some G-force especially once it takes off..It kind of reminds me of being in a roller coaster or the gravitron ride.......It's very smooth after it takes off and once it reaches a certain height in the sky you actually forget that your flying. It's Fun!!
