it's kind of like saying uncle, or mister here. Kun is something used as a familliar term after a boy's name. His close friend or parents would call him Jim-kun. For girls or something cute it would be chan. Jen-chan. This is familliar though, don't use it unless you know the kid well, and I'd say avoid using it with adults altogether.

Sama is a formal term of respect for a superior Smith-sama.

San a general, every day term of respect, usable by anyone (like when you're addressing your teacher), and attached after the last name. Smith-san (this one is most like Mr, but it can be used for things other than people.)

it's mostly to differentiate seniority between your friends

kun = if u had a little Brother named naruto...u would call her alice-kun ...coz he is your younger brother

sama = refers to the head or leader

san = refers to Mr or Mrs e.g. Alice-san / Ben-san
