Neither would be good.

First -- it is "grammar" -- not "grammer."

Second -- it is "ad" -- not "add" -- and you would be better to use 'advertisement."

Third -- they know you are writing to them -- why do you have to tell them?

Fourth -- if you insist on this, I'd write "I am writing regarding your advertisement . . . ."

When you are done with it -- use both Spell-Check and Grammar-Check on it.

I would actually go with...."I have a question regarding an advertisement..." (or "ad", not "add")

It is not necessary to explain that you are writing. They will have figured that out once they have your letter in their hands.

*edit* Better yet, don't tell them "what you are doing". Just ask the question....

"You have an advertisement for *widget*. What sizes are available.....?"

Er... do you mean "ad"? Short for "advertisement"?
You could say "as regards", "in this (or that) regard", or "with (or in or having) regard to".

Also, "grammar".

"I am writing regarding an ad" is the least clumsy. (Note spelling of "ad".)

ad is short for advertisment, not add.

you should spell it out if your are writing a formal letter.
