Well there are many ways to do that, it depends what string and what octave you want to play. You can play an E in any of the following ways:
open low E string
12th fret on low E string
7th fret on A string
19th fret on A string
2nd fret on D string
14th fret on D string
9th fret on G string
21st fret on G string (if your guitar has enough frets)
5th fret on B string
17th fret on B string
open high E string
12 fret on high E string
Alright, that's just about every E note on the neck of the guitar, (unless you have an electric with 24 or more frets).

quite simple, the thinnest and fattest strings on the guitar (assuming it is tuned correctly AND it is a non-classical six string) are tuned to E.
Now, on the second fattest string (which should be tuned to A) the seventh fret down is also e. there are many E's on the guitar.
check out sites like chordfind.com and others to help you form chords and even tune.

you pluck the 1st or 6th string unfretted.. high e or low e.. but if you experiment with a piano and guitar im sure you will find the note your lookin for..

The top string is low e and the bottom string is high e. Pluck it without holding it down.
