The best way to make a unique backpack is to make it yourself, or to put your personal touch on it.
I bought a very simple black, corduroy backpack from PacSun during my sophmore year in high school (3 years ago). It had 3 tiny patches on it.
I took it from there, and since I travel a lot, I bought 1 patch in every country that I went to (Greece, Italy, Spain, The Netherlands, etc). Then I just sewed them on. Well not really sewed. I used a needle in thread in such a way that I managed to keep the patches on the backpack and still make it look neat ;)
The finished product looks kind of like those travel trunks that you see in old cartoons that have the stickers pasted all over them.
I get SO MUCH response to my backpack. People who I don't even know ask me where I got it!
You could also look into duct tape backpack. Duct tape is very durable and easy to craft things out of. Right now, I'm making a duct tape purse that holds my iPod and has a headphone jack in it, so I can plug my headphones into the purse itself and it plays the music from my iPod. It's going to look like an old school boombox when I finish it.
The best way to get a unique backpack is to come up with a unique idea and go with it.

Check out eBay, look up backpacks and check under vintage. Or if you're willing to lay down some cash, check out the Apple (computers) store. They have laptop bags that double as backpacks in all styles, that are stylish whether you actually have a laptop or not. No one will know the difference, they're made to protect whats inside, and if you have (or later get) a laptop, you can carry it AND all your stuff in there.

i had a military back pack i used when i was riding my motorcycle, it was very durable, i took it to New Mexico and California and all over Arizona, it held up great until i was t-boned on my bike and the ambulance crew cut the straps to take it off my back. it is also water resistant, i was rained on many times and my stuff stayed dried. i am a big person and the straps adjusted to fit me,,and they did not come loose because they were stretched out to the max length. that was a damn good back pack,
i want another one myself, so off to the near by Army surplus store to get one, they have many varieties to choose from. but they are military OD green or camouflage. if there is a military base by you, you will not have any problem getting one. they might be a little pricey, but well worth it.

military surplus rucksack. not a lot of people have them and they last for years of hard abuse when normal school bags only last a few months carrying papers.
