No, Toad cameos as Peach's shield by holding down + B. He also appears in adventure mode on several instances in Level 1's vast bacckdrop. He and Koopa Troopa and Paratroopa appear as trophies.
If by "Koopa" you mean "Bowser" you are retarded. If by "Toad" you accidentally meant "Wart" the final boss who is a giant toad in SMB2, he appears as a trophy as well.

Most people do realize that Koopa *is* Bowser. Those who don't should be hit with a Boot.
But, no. Toad is Peach's block. Toad isn't unlockable. Neither is Sonic.
