Nobody is supposed to know. It wasn't really confronted because they couldn't really decide on what it should be. At the last minute it was suggested by someone on the set that they throw in an orange lightbulb to give in an unearthly glow, which has led to speculation that it is either gold, or Marsellus Wallace's soul. The latter of which is the most popular, and leads of course to more speculation that Marsellus made a deal with the devil and the college kids that Vincent and Jules shot were really followers of Satan, or lesser Demons, (which explained in many people's minds why Jules' insisted that the bullets that missed them were a result of divine intervention) and that the band aid on the back of Marsellus' neck was actually hiding the mark where Satan sucked out his soul.
This is of course just baseless trivia. Tarantino likely didn't intend that much backstory for the suitcase, it was just there as a plot device, and the viewers drew some pretty wild conclusions based on something done on a whim.

I just watched that and was wondering the same thing. I don't think they show it. The yellow glow makes it look like it is gold but then the suit case should have wieghed a bunch.

My guess: Gold. The colored reflection on John's and Samuel's skin was definatly gold.

Man, I've always wondered that myself!!! I think maybe it was gold or something. It looked like it was shiney. Couldn't have been drugs or money. Hope someone answers who knows!
