The "frankly my dear, I don't give a...." scene - Gone with the Wind
The "just whistle" scene - Bogy and Bacall

Man, I just love movies. Especially the classics. I could probably
list 100, but those are 2 of my faves.

EDIT:
"You can't handle the truth" - A Few Good Men
Quint talking about the sharks eyes - Jaws
The rolling ball scene - Raiders of the Lost Ark
The final scene where the ex governor walks in only to find his ex slave is now governor - Captain Blood
The final scene where they read the wire saying how much money will be sent and "Daddy everytime a bell rings, an angel gets his wings" - It's a Wonderful Life
The head being smashed - The Untouchables
John Wayne's character saying how many broken parts he's had, but can still beat the crap out of Bruce Dern's character - The Cowboys
The scene where the doc tells Books that he's going to die - The Shootist
The scene where they shove the needle in her chest and she gets up - Pulp Fiction
"snakes, why did it have to be snakes?" - Raiders of the Lost Ark

I love that scene in Big. In fact, I learned that FAO Schwartz made & sold 3,000 of those pianos.

Getting back to your question, here are a few others:
- "This is the beginning of a beautiful friendship" (Casablanca)
- Rocky running up the steps at that Philadelphia landmark
- Dorothy opening the door to Munchkin land (first color scene in a movie
- Singin' in the Rain (i.e. Gene Kelly dancing to it)
- "Show me the money" and "You had me at hello" (Jerry Macguire)
- The horse head scene from the Godfather (yuck!)
- The shower scene in Pyscho
- "If you build it, they will come" (Field of Dreams)
- "Houston, we have a problem" and "Failure is not an option" (Apollo 13)
- "Despite everything, I still believe people are good at heart" (Diary of Anne Frank)
- Thelma & Louise driving over/into the Grand Canyon
- "Shall we play a game" and the whole tic-tac-toe/Global Thermo Nuclear war game (War Games)
- John Travolta dancing in Saturday Night Fever
- Summer Lovin' from Grease
- The Russian Roulette scene from The Deer Hunter
- When Meryl Streep has to make the choice in Sophie's Choice
- I had him for dinner with a nice Chianti, followed by hissing sounds from Silence of the lambs
- The orgasm scene from When Harry Met Sally
