Yes. If you want to go the non-cheating route, all you have to do is go out at night (either buy a car or take a cab) to a downtown lot. If you're lucky, a grand vampire count/countess will be hanging around. Go over and chat them up for a bit, and try and make friends. it'll take a while, since they have the annoying habit of running away at sunrise. Keep inviting them over to your house at night, or see if you can catch them on a lot.
After a while, when your daily/lifetime relationship scores are around 50 or so (or when you see the little "friend" smiley), invite them over to your house for a little get together. If you're nice to them, and they like you enough, you'll get bitten in no time :)
Of course, I'm lazy, so I download hacked objects and mods that make it easier for my simmies to go vampy :P

Yes, you can. I had a female sim make friends with the male vampire. Eventually he bit her and turned her into a vampire. Then I had her go home and turn her husband and son into vampires too!
I hear that in the new Pets expansion pack, you can turn your sims into werewolves! That would be pretty cool too.
