It's a threat. Can you trust him? If he tells, just act cool & say "That's not true" to them. It will seem that you're more credible than him for them to believe.

don't be worried about it. he's either calling a bluff, he's not serious about telling, or he's a jacka$$.
no matter what he's doing, you don't have to put up with it. if you want to, you could simply refuse to clean his room (although you should "clean" it, in order to find stuff about him :-) and threaten to tell whoever has authority over him that he was blackmailing you. (you'll be just as bad as him, but at least you'll have an upper hand.)
if he does tell on you, just laugh it off and tell everyone that it is completely false. if you act like the whole thing is unimportant, people will believe you and not him.
if you want to be really horrible to him, you could always just say that the reason he spreads these rumors around is because he doesn't want anyone to know he has herpes or something.

Honestly, does it really matter if your crush knows you like him? Seriously, it's not like you going to get married.
So here are your options.
1. Your brother tells, your crush tells him that he likes you too. You go to a movie.
2. You brother tells him and he doesn't like you. He moves on. You move on.
3. Your brother squeals, crush doesn't like you in 'that way' but woudn't mind being friends. You have new friend, brother is now known as a 'snitch'.
4. You clean your bro's room, he tells anyway, insert one of the above results here.
5. You clean room, bro keeps his trap shut, crush finds new girl, you are sitting home alone, in your brother's clean room.
6. You tell crush, by dropping subtle hints that you like him. Bro's room remains bro's problem. You and crush go to dinner & a movie.
Yeah, so what's what the question again?

You can't.
So make the most of the situation and turn your crush into a relationship.
The more people who think you are serious the better and your brother will look the fool.

stop trying to hide whatever it is you are hiding if you feel that you have to hind it then you must also feel that it is the wrong thing to be doing in the first place. If you stop hiding it then the blackmail could not happen

There is only one way to avoid being blackmailed. Stop being threatened by it. If you act worried, he will always find ways to blackmail you.

You could always lie about it and you don't have to give in to what he wants. What's the worse can happen if he actually tells everybody?
