Normally one day, or two at the most. But to be sure, contact the post office and ask them what time is their next pick up, and they will tell you what time their pick up is.If you have something to mail, for instance, an important letter, the post office pick up times are 2:00 and 4:00 pm, you can mail the letter an 1/2 hour before pick up time.
That's how I do it. I just call them and ask about their pick up times throughout the week. Normally they keep the same schedule, but sometimes it can change...

It all depends on when and where you mail the letter and how fast or efficient the mail system is. Now sometimes if you mail a letter and place it in the mailbox on a Saturday night, certain places will take that mail out and mail it. Other places will let it sit in the box until the following Monday, it all depends.
It also depends on if there is a holiday or some day that the postal people have a day off could deter the mail to get to it's destination in proper amount of time.
Each town and city operates differently. It just all depends. It could be the same day, the next day or two days later.

I live in Western Mass, and things take 2 days to get to Boston area, northern Connecticut, or southern Vermon, but usually 1 day to get to another Western Mass town. For me, most places outside of New England on the East Coast take 3-4 days. (Florida for example). When I sent letters to the West Coast it takes 5-7 days.
Depending upon how large your "city" is, 1-2 days is all it should take for transit, tops. If you're really concerned, pay the extra $2.40 or whatever for certified letter. Good luck with that! :)
