On the evening of our parents wedding my brother and stepsister went of to screw while his wife looked after his 2 kids. Should I tell his wife? Confront him? Our step sister was the one who told me about the fling. Personally I think it's wrong to screw around let alone with your step sister!

You shouldn't tell. It all will come back on you will be accused of causing family strife. What is done in the dark will come to light eventually and you don't have to deal with anyone who will blame you.
ps I also agree with Brianna thats an excellent idea to confront him and let him telll his wife

I dunno about you but every time I have known someone who did this and I said anything to anyone about it..... i ended uup being the bad guy...
Tell your brother you know... Tell your brother your step sister told you... tell him how you feel about... leave it all right where it belongs.... in his lap... It is not you mess clean up... it is his...
If his wife never finds out.... then she never finds out... if she does... you do not want to be in the midst of it all.

Hell, no. Don't tell. Instead, you should consider them secret lovers, help them to keep it a secret, because they may really have feelings for each other. Did you know that years ago, brothers and sisters, unkles and aunts, etc, were married and had children?
