What's up comes from New York City. Around the turn of the 20th Century people were entering Ellis Island and the first thing they saw was the Statue of Liberty. The Captain would motion to the passengers to look and in one of these voyages a African American worker coming from North Carolina to drop off a new boat look at the statue and said "Now that's wussup!" So it went from there.

From Bugs Bunny's habit of saying "What's up, Doc?" But I don't know where he got it; maybe it was not original with him. However, Bugs is pretty old, so maybe it WAS original with him.

I'm not totally sure about this but i think it had something to do with a sexual inuendo "What's Up?" (think about it)
