True course is the direction of the line connecting two desired points, drawn on the chart and measured clockwise in degrees from true north on the meridian.

Draw a line between your two points on your sectional then use your course plotter to take its direction off one of the meridians or parallels which are printed on the chart. By using the meridians, TC directions from one point to another are be measured in degrees, in a clockwise direction from true north. That's your true course.

In addition to what av8trx said, once you find the true course, you need to add or subtract the wind correction angle. This is done using your flight computer.

True course +/- wind correction angle = true heading
True heading +/- magnetic variation = magnetic heading
Magnetic heading +/- magnetic deviation = compass heading
