Amtrack will get you from Buffalo (either Bflo downtown or Depew) in about 8 hours and 45 minutes for $79.
Greyhound bus will get you there in a little over 8 hours for $78

Buffalo to NYC?
I'd say drive, it'll be long, but you know.
If you take a plane, it'll probably be like a 30 minute to an hour ride. If you take a train, I'd say 3 hours.
Driving will be long and maybe dangerous. Go on mapquest and see how long the time and distance is from Buffalo to NYC.

I would suggest Amtrak. Its probably the most worry free way to make that trip since it offers the many comforts of a plane but without the hassle of airport security. You might enjoy a nice nap or a drink contrary to driving up.

The fastest way would be by plane. If you drive, you're looking at over six hours to get there, and by bus it will probably take ten because you have to stop often. If you have to drive, take the thruway; it's always the most direct and safest route to get anywhere in New York.

Plane or Train to Penn Station. You could also get a bus. Not sure how long the drive would be though.

I would travel by train - not as fast as a plane - but beautiful, scenic views.
If you truly need to get there fast, then a plane is the only way.
