Jesus, being one manifestation of the triune God, was active in the creation of the world as we know it. In this sense, He is the root of all things, including king David.

He can also be considered the offspring of David, as both his earthly mother and her husband were physically decended from the ancestry of king David.

By root, He meant that He is the center of all life. It's like saying that He is the root of the tree of life, and we are all the branches and leaves that grow from Him. Without Him, we die. If you cut off a branch or a leaf, it falls to the ground and dies. Therefore, you must stay connected and firmly rooted in Jesus and you will have life.
