Give a list of short chores. He's only 5.
If you say "clean your room" it may seem overwhelming for him to mentally organize all of the tasks and he will have trouble understanding what you consider a mess.
Show him how to put away categories of things: hang up clothes; put away all books; put away all games; put away all sports equipment (like that). Empty trashcan.
Teach him how to make a bed, but don't expect perfection.
Praise each step along the way.
Make sure he has enough storage space and shelving.
5-yr. olds are usually pretty easy to work with.
Remember to break down the assignment of cleaning into smaller bits so they understand the process. :)
And no TV or play time til the job is done! Be firm.
My daughter is teaching her toddlers to clean rooms now and they are pretty good at it. Stay consistent.

In order to get a 5 year old to clean his bedroom you need to show him how much "fun" it is to clean your room. Try reverse psychology, let him know that cleaning up your room can be as much fun as playing video games. If he likes Barney for instance, try the clean up song. Make it fun.

You could try an award jar. You could put money in there for everytime he cleans it. You could also write down on a piece of paper something like Ice Cream/Movie/ETC and if he has cleaned his room for a week straight with no problems then have him choose something from the jar.
Don't let him watch tv/play games/ETC until he has cleaned his room.

Take away all his stuff. Every single thing he has that makes his room 'dirty'. Then there is nothing to clean up. If you give your kid nothing to make a mess with, there is no mess.
For clothing-a trick the works is to get a set or two of ulgy clothing. Anything he does not like. A pick shirt or a salior shirt, whatever. Then you keep thoes clothes washed and ready for him to wear. If he wants his good clothes washed, he has to put them in the hamper(or whatever) himself. You might have to get rid of a favorate piece of clothing, as he may try to wear it 'dirty' for a couple days.

Tell him why he needs to clean his room. But you have to start training him with little things like keeping his toys, keeping his books etc after using it. Explain why. Sometimes it works when we tell the effect of what they are doing, when you keep a toy you'll know where to get it when needed. You help mommy clean the house..etc. Praise him for a job well done. When i acknowledge my kid's accomplishment he likes it that he is appreciated.

Remind him that this bedroom is in your house thus making it your room that you are kind enough to let him stay in and the rule is he is to keep that room neat and tidy.

let him know who the parent is. why is it an option for him NOT to clean his room?
does he know how? if not start small and praise good efforts, if he refuses, time out, no chocolate.
well, the chocolate works for my kids.
