"You shouldn't give up on him" is correct. It means that you should trust him longer and have patience with him. The second sentence you wrote has no meaning. There is another similar sentence which has a different meaning: "You shouldn't give him up" implies that you have him (maybe he is your boyfriend or your employee) and you should not get rid of him.
