I the person's first language is not English, then I would ask a second time because, in my personal experience I have said yes but really meant no, not at all.

No, of course, not. If you were going to just do it anyway, why bother to ask? Once you have asked, doing it when they have said it bothers them is an insult to them -- it says you have no regard for their feelings in the matter, despite having pretended to by asking. Why not just slap them?
If you want to do something *that* badly, just don't ask -- and then deal with the consequences that arise.

why would you ask them if the mind if you are going to ignore the response to the question?
Personally I would be offended if you asked me that and I said no and you went ahead and did it anyway. What is the point of asking?

Absolutely not. If their answer didn't matter to you, you shouldn't have asked in the first place. To do something after someone's told you it bothers them is even more rude than just doing it in the first place.
