It takes more than a thumb and it is against an artery, temporarily blocking blood from the brain long enough for the brain to shut down. If you really need to know this for self-defense, take a class to learn that technique. It can be dangerous to attempt it without instruction, as it can damage a "subject's" windpipe (esophagus) if done incorrectly.

I don't know why you want to know this, but here goes:
If the thumb is placed correctly, directly below the nose, at the base of the neck, near the dip in the breast bone, and a large amount of pressure is placed on the neck right at that point, you can cut off all air goint to the lungs, brain, and heart. This will cause the person to pass out. Any higher or in a different area will cause the person to vomit. A extremely lage amount of pressure placed on the neck where ever a pulse is strong (very strong) will cause a vein to rupture, killing a person.
Please! Never use any of this information to harm anyone.

Place your thumbs on both sides of the throat until you find their pulse (the carotid).

Once you are sure that you can feel both pulses, apply a small amount of pressure.

The recipient should start filling a bit faint.. don't do this for too long, you don't want to cut the blood supply to their brain for too long.

you have to block their jugular artery, so blood stops flowing to the brain and they pass out...(the jugular is the tube thing on the side of your neck, push it down long enough, they'll go unconscious)
